1  Scope
This International Standard describes a method for the determination of soluble nitrogen fractions (nitrate, nitrite,
ammonium and organic nitrogen) in a 0,01 mol/l calcium chloride extract of soil samples.
It is applicable to air-dry soil samples, pretreated in accordance with ISO 11464.

3  Principle
By means of a simple extraction with a 0,01 mol/l calcium chloride (CaCl ) solution of air-dry soil samples, different

soluble nitrogen fractions are extracted (see references [1] and [2]). The concentrations of the inorganic nitrogen
compounds nitrate (+ nitrite) and ammonia are measured directly in the extract using automated spectrometric
methods.
For the determination of total soluble nitrogen, part of the extract is first digested whereafter the produced ammonia
is converted to nitrate. This, together with the originally present nitrate (+ nitrite) and ammonia, is then measured
using an automated spectrometric method. The content of soluble organic nitrogen is calculated by subtracting the
content of nitrate (+ nitrite) and ammonium nitrogen from the content of soluble total nitrogen.
NOTES
1  The content of soluble organic nitrogen may give an indication of the easily mineralizable fraction of the organic matter.
2  The content of extractable ammonium and organic nitrogen of the soil is changed very often by drying, compared to the
fresh soil sample. The way of drying affects the results (see reference [3]).
3  Since manual determinations for the different nitrogen fractions cause problems, in this International Standard
measurements are specified using automated continuous flow techniques. Other ways of detection are permissible provided
the results are in agreement with the results achieved by the specified automated procedures.

4  Laboratory samples
The fraction of the air-dry soil samples, pretreated in accordance with ISO 11464, which has passed a sieve of
2 mm aperture shall be used. Part of this sample shall be used to determine the water content in accordance with
ISO 11465.
5  Extraction
5.1  Principle
Air-dry soil is extracted with a 0,01 mol/l calcium chloride (CaCl ) solution in a 1:10 ratio (m/V) at 20 °C – 1 °C. After

reaching equilibrium (2 h), the solution is centrifuged for the determination of the different nitrogen fractions.
NOTES
1  The extraction is performed at a constant temperature of 20 °C because the amounts of extractable ammonium nitrogen and
soluble organic nitrogen are affected by the temperature of the extracting solution (see reference [4]). For special climatic
conditions, another constant temperature may be chosen. The used extraction temperature should be mentioned in the test
report.
2  Centrifuging is preferred because most filter papers either absorb substances or may be contaminated by ammonia. When
the extracts are to be filtered, the filter papers should be stored in a desiccator over concentrated sulfuric acid or a drying
medium containing sulfuric acid, one week at least before use, and the first 20 ml of the filtrate should be discarded.
5.2  Reagents
Use only reagents of recognized analytical grade.
5.2.1  Water, with a specific conductivity not higher than 0,2 mS/m at 25 °C (water according to grade 2 of
ISO 3696).
5.2.2  Calcium chloride solution, c(CaCl ) = 0,01 mol/l, for the extraction.

Dissolve 1,47 g of calcium chloride dihydrate (CaCl � 2H O) in water (5.2.1). Dilute with water to make 1 litre.

NOTE —  CaCl � 2H O may absorb water on standing. The reagent should be standardized.

5.3  Apparatus and glassware
5.3.1  Analytical balance, with an accuracy of 10 mg.
5.3.2  Polyethylene bottles, of nominal volume 250 ml, with screwcaps.
5.3.3  Shaking machine or reciprocating shaker, 150 r/min to 250 r/min.
5.3.4  Centrifuge, capable of holding the tubes used.
5.3.5  Polyethylene centrifuge tubes, of nominal volume 100 ml or other sufficient volume.
5.4  Procedure
Weigh 10,00 g of the laboratory sample into a polythene bottle (5.3.2).
Add 100,0 ml of calcium chloride solution (5.2.2) at a temperature of 20 °C. Shake mechanically (5.3.3) for 2 h at
20 °C.
Decant the necessary quantity of the extract suspension into centrifuge tubes (5.3.5) and centrifuge (5.3.4) for
10 min at about 3 000 g. Decant the supernatant solution in measuring cups and measure the contents of nitrate
(+ nitrite), nitrite (if necessary), ammonium and total soluble nitrogen, as described in clauses 6, 7, 8 and 9
respectively.

Perform a blank test by adding only the calcium chloride solution (5.2.2) to the polythene bottle (5.3.2).
The soluble nitrogen fractions should be measured immediately, but not later than one day, after the extraction. If
this is impossible, the extracts should be stored in a refrigerator at temperatures not exceeding 4 °C, for at
maximum one week.
6  Determination of nitrate nitrogen
6.1  Principle
In a segmented flow analysis (SFA) system, the sample is first subjected to dialysis. Nitrate and nitrite ions from the
samples pass in the membrane and are taken in an understream of ammonium chloride. The nitrate is then reduced
to nitrite by means of cadmium. Next, a-naphthylethylenediamine dihydrochloride and sulfanilamide are added, so
that a red-coloured diazo compound is formed in the acid medium. Its absorbance is measured at a wavelength of
543 nm.
NOTES
1  The dialysis serves to separate nitrate ions from interfering substances such as solid particles, colloids and coloured
organic compounds.
2  The colour reagent is known as the Griess-llosvay reagent.
Normally, the content of nitrite in soils is negligibly small. In these cases, the sum of nitrate and nitrite corresponds
to the content of nitrate. In special cases, when nitrification in the soils seems to be incomplete, the content of nitrite
has to be determined separately.
6.2  Reagents
6.2.1  Wetting agent, polyoxyethylene lauryl ether (30 %).
NOTE —  The wetting agent solution should not be stored for longer than 6 months.
6.2.2  Buffer solution
Dissolve, in a 1 litre volumetric flask, 25 g of ammonium chloride (NH CI) in a little water. Add 12,5 ml of ammonium

hydroxide solution [w(NH OH) = 3 %] and 1 ml of wetting agent (6.2.1). Make up to the mark with water and mix.

The pH of this solution should be between pH 6 and pH 8.
6.2.3  Cd/Cu reducing agent
Swirl approximately 5 g of cadmium powder (particle size 0,3 mm to 0,8 mm) for 1 min with about 30 ml of
hydrochloric acid [ (HCl) 1 mol/l]. Wash with water until acid free. Then add about 50 ml of a copper (II) sulfate
c =
solution [r(CuSO = 20 g/l] and swirl for 3 min. Wash at least 10 times with water to remove any flocculated copper.

Store the Cd/Cu reducing agent in a dark place.
6.2.4  Cd/Cu reduction tube
Fill the U-shaped column (6.3.2) with buffer solution (6.2.2), taking care not to introduce air bubbles. Introduce the
activated cadmium powder (6.2.3) with the aid of a funnel on both sides of the column. Apply vibration now and then
to pack the powder. Fill the column up to 5 mm from the top of the column and seal the ends with a small plug of
glass wool.
The column is now ready for use and can be placed in the SFA system.
NOTE —  To lengthen the lifetime of the Cd/Cu reducing agent, a copper wire should be installed in the beginning of the
Cd/Cu reduction tube, and renewed if necessary.

6.2.5  Colour reagent
In a 1 litre volumetric flask, add 150 ml of concentrated phosphoric acid [w(H PO ) = 85 %] to 0,5 l of water. Add

0,5 g of a-naphthylethylenediamine dihydrochloride (C H N Cl ) and swirl until dissolved. Then dissolve 10 g of

sulfanilamide (C H N O S) in this mixture and make up to the mark with water.

6.3  Apparatus
6.3.1  Segmented flow analysis system
The system consists of a sampler, pump, dialysis unit, reduction column, nitrate unit, photometer and recorder. The
flow diagram of an appropriate segmented flow analysis (SFA) system is given in annex A (figure A.1). Note that
these flow diagrams may require adaptation, depending on the system used.
6.3.2  Cd/Cu reduction tube
This comprises U-shaped glass tubing, about 15 cm long and with internal diameter of 2 mm, provided with ferrules
for connection to the SFA tubing. It may be purchased filled with cadmium from the SFA system manufacturer, or be
home-made (see 6.2.3 and 6.2.4).
NOTE —  This SFA arrangement allows determination of nitrite by simply shortcutting the reduction column (see annex A,
figure A.2).

L'ISO (Organisation internationale de normalisation) est une fédération
mondiale d'organismes nationaux de normalisation (comités membres de
l'ISO). L'élaboration des Normes internationales est en général confiée aux
comités techniques de l'ISO. Chaque comité membre intéressé par une
étude a le droit de faire partie du comité technique créé à cet effet. Les
organisations internationales, gouvernementales et non gouvernementales,
en liaison avec l'ISO participent également aux travaux. L'ISO collabore
étroitement avec la Commission électrotechnique internationale (CEI) en
ce qui concerne la normalisation électrotechnique.
Les projets de Normes internationales adoptés par les comités techniques

Qualité du sol — Détermination de l'azote nitrique, de l'azote
ammoniacal et de l'azote soluble total dans les sols séchés à l'air
en utilisant le chlorure de calcium comme solution d'extraction
1  Domaine d'application
La présente Norme internationale décrit une méthode de détermination des fractions d'azote soluble (nitrate, nitrite,
ammonium, azote organique) dans un extrait d'échantillon de sol par du chlorure de calcium à 0,01 mol/l.
La présente Norme internationale est applicable aux échantillons de sol séchés à l'air et traités préalablement
conformément à l'ISO 11464.
2  Références normatives
Les normes suivantes contiennent des dispositions qui, par suite de la référence qui en est faite, constituent des
dispositions valables pour la présente Norme internationale. Au moment de la publication, les éditions indiquées
étaient en vigueur. Toute norme est sujette à révision et les parties prenantes des accords fondés sur la présente
Norme internationale sont invitées à rechercher la possibilité d'appliquer les éditions les plus récentes des normes
indiquées ci-après. Les membres de la CEI et de l'ISO possèdent le registre des Normes internationales en vigueur
à un moment donné.

3  Principe
Différentes fractions d'azote soluble sont extraites par simple agitation d'échantillons de sol séchés à l'air dans une
solution de chlorure de calcium (CaCl ) à 0,01 mol/l (voir références [1] et [2]). Les concentrations des composés

d'azote inorganique, nitrate (+ nitrite) et ammonium sont mesurées directement dans l'extrait à l'aide d'une méthode
spectrométrique automatisée.
Pour la détermination de l'azote soluble total, une partie de l'extrait est mise en digestion, puis l'ammonium résultant
est transformé en nitrate. Ce nitrate, ainsi que le nitrate initial (+ nitrite) et l'ammonium sont alors mesurés à l'aide
de la méthode spectrométrique automatique. Pour calculer la teneur en azote organique soluble, on soustrait la
teneur en nitrate (+ nitrite) et en azote ammoniacal de la teneur en azote soluble total.

NOTES
1  La teneur en azote organique soluble peut indiquer la fraction des matières organiques faciles à minéraliser.
2  La teneur en azote ammoniacal et en azote organique extractibles du sol est très souvent différente de celle de l'échantillon
du sol de départ à cause du séchage. La façon de sécher influence les résultats (voir référence [3]).
3  Comme les déterminations manuelles des différentes fractions d'azote causent des problèmes, les mesurages spécifiés
dans la présente Norme internationale sont effectués à l'aide de techniques automatisées de flux continu. D'autres méthodes
de détection sont admises à condition que les résultats coïncident avec ceux obtenus par des modes opératoires automatiques
spécifiés.
4  Échantillon pour essai
Utiliser la fraction tamisée à 2 mm des échantillons de sol séchés à l'air, préalablement traités conformément à
l'ISO 11464. Utiliser une partie de l'échantillon pour déterminer la teneur en eau conformément à l'ISO 11465.
5  Extraction
5.1  Principe
Le sol séché à l'air est agité dans une solution de chlorure de calcium (CaCl ) à 0,01 mol/l dans un rapport de

1:10 (M/V) à 20 °C – 1 °C. Après avoir atteint son équilibre (2 h), la solution est centrifugée pour la détermination
des différentes fractions d'azote.
NOTES
1  L'extraction est effectuée à une température constante de 20 °C car les quantités d'azote ammoniacal et d'azote organique
soluble extractibles varient en fonction de la température de la solution d'extraction (voir référence [4]). En cas de conditions
atmosphériques particulières, une autre température constante peut être choisie. Il convient de mentionner dans le rapport
d'essai la température d'extraction choisie.
2  La centrifugation est choisie de préférence, car la plupart des papiers-filtres absorbent les substances ou peuvent être
contaminés par l'ammoniac. Lorsque les extraits doivent être filtrés, il convient d'entreposer les papiers-filtres, une semaine au
moins avant leur utilisation, dans un dessiccateur sur de l'acide sulfurique concentré ou sur un support sec contenant de l'acide
sulfurique, et il convient de jeter les premiers 20 ml du filtrat.
5.2  Réactifs
Utiliser uniquement des réactifs d'une qualité analytique reconnue.
5.2.1  Eau, d'une conductivité spécifique ne dépassant pas 0,2 mS/m à 25 °C (eau conforme à la qualité 2 de
l'ISO 3696).
5.2.2  Solution de chlorure de calcium, c(CaCl ) = 0,01 mol/l, pour l'extraction.

Dissoudre 1,47 g de chlorure de calcium dihydraté (CaCl ,2H O) dans de l'eau (5.2.1). Diluer avec de l'eau jusqu'à

obtention de 1 litre de solution.
NOTE —  CaCl ,2H O peut absorber de l'eau lors du stockage. Il convient de vérifier la composition du réactif.

5.3  Appareillage et verrerie
5.3.1  Balance analytique, d'une précision de 10 mg.
5.3.2  Flacons en polyéthylène, d'un volume nominal de 250 ml, dotés de couvercles à vis.
5.3.3  Agitateur à va-et-vient, 150 r/min à 250 r/min.
5.3.4  Centrifugeuse, capable de contenir les tubes utilisés.

5.3.5  Tubes à centrifuger en polyéthylène, d'un volume nominal de 100 ml ou d'un autre volume suffisant.
5.4  Mode opératoire
Introduire 10,00 g de l'échantillon pour essai dans un flacon en polyéthylène (5.3.2).
Ajouter 100,0 ml de la solution de chlorure de calcium (5.2.2) à une température de 20 °C. Agiter à l'aide de
l'agitateur (5.3.3) pendant 2 h à 20 °C.
Transvaser la quantité nécessaire de la suspension extraite dans des tubes à centrifuger (5.3.5) et centrifuger
(5.3.4) pendant 10 min à environ 3 000 g. Transférer le surnageant dans des tubes à hémolyse et mesurer les
teneurs en nitrate (+ nitrite), nitrite (si nécessaire), ammonium et en azote soluble total, comme décrit aux articles 6,
7, 8 et 9, respectivement.
Effectuer un essai à blanc en ajoutant seulement la solution de chlorure de calcium (5.2.2) dans le flacon en
polyéthylène (5.3.2).
Il convient de mesurer les fractions d'azote soluble immédiatement, mais pas plus tard qu'un jour, après l'extraction.
Si cela n'est pas possible, il convient de conserver les extraits dans un réfrigérateur à une température ne
dépassant pas 4 °C, pendant une semaine au maximum.
6  Détermination de l'azote nitrique
6.1  Principe
Dans le système à flux segmenté (SFS), l'échantillon est d'abord dialysé. Les ions nitrate et nitrite traversent la
membrane et passent dans un flux de chlorure d'ammonium. Le nitrate est ensuite réduit en nitrite à l'aide de
cadmium. De la dihydrochlorure de a-naphtyléthylènediamine et du sulfanilamide sont ensuite ajoutés, de sorte
qu'un composant diazo de couleur rouge soit formé en milieu acide. Son absorbance est mesurée à une longueur
d'onde de 543 nm.
NOTES
1  La dialyse sert à séparer les ions nitrate des substances interférentes, telles que les particules solides, les colloïdes et les
composés organiques colorés.
2  Le réactif de coloration est connu sous le nom de réactif de Griess-Ilosvay.
Normalement, la teneur en nitrite des sols est quasiment négligeable. Dans ce cas, la somme de nitrate et de nitrite
correspond à la teneur en nitrate. Dans certains cas, quand la nitrification dans le sol semble incomplète, la teneur
en nitrite doit être déterminée séparément.
6.2  Réactifs
6.2.1  Agent mouillant, éther laurique de polyoxyéthylène (30 %)
NOTE —  La solution d'agent mouillant ne doit pas être conservée au-delà de six mois.
6.2.2  Solution tampon
Dans une fiole jaugée de 1 000 ml, dissoudre 25 g de chlorure d'ammonium (NH Cl) dans une petite quantité d'eau.

Ajouter 12,5 ml d'une solution ammoniacale [w(NH OH) = 3 %] et 1 ml d'agent mouillant (6.2.1). Ajouter de l'eau

jusqu'au trait et mélanger.
Le pH de cette solution doit être compris entre 6 et 8.
6.2.3  Agent de réduction Cd/Cu
Mélanger énergiquement environ 5 g de poudre de cadmium (taille des particules comprise entre 0,3 mm et
0,8 mm) pendant 1 min avec environ 30 ml d'acide chlorhydrique [c(HCl = 1 mol/l)]. Rincer avec de l'eau jusqu'à

élimination totale de l'acide. Ajouter ensuite environ 50 ml d'une solution de sulfate de cuivre (II) [r(CuSO = 20 g/l)]

et mélanger pendant 3 min. Laver au moins 10 fois avec de l'eau pour retirer tout précipité du cuivre. Conserver à
l'obscurité l'agent de réduction Cd/Cu.
6.2.4  Tube de réduction du Cd/Cu
Remplir la colonne en forme de U (6.3.2) avec la solution tampon (6.2.2), en veillant à ne pas laisser pénétrer des
bulles d'air. À l'aide d'un entonnoir, verser la poudre de cadmium activée des deux côtés de la colonne. Secouer de
temps en temps pour tasser la poudre. Remplir la colonne en laissant un espace de 5 mm aux extrémités et
boucher celles-ci avec de la laine de verre.
La colonne est alors prête à être utilisée et peut être placée dans le système SFS.
NOTE —  Pour allonger la durée de vie de l'agent de réduction Cd/Cu, il convient de placer un fil de cuivre à l'entrée du tube
de réduction Cd/Cu, et de le renouveler si nécessaire.
6.2.5  Réactif de coloration
Dans une fiole jaugée de 1 000 ml, ajouter 150 ml d'acide phosphorique concentré [w(H O P) = 85 %] à 0,5 l d'eau.

Ajouter 0,5 g de dihydrochlorure de a-naphtyléthylènediamine (C H N Cl ) et mélanger jusqu'à dissolution.

Dissoudre ensuite 10 g de sulfanilamide (C H N O S) dans ce mélange et compléter avec de l'eau jusqu'au trait.

6.3  Appareillage
6.3.1  Système d'analyse à flux segmenté
Le système se compose d'un échantillonneur, d'une pompe, d'un dialyseur, d'une colonne de réduction, d'une unité
pour nitrate, d'un photomètre et d'un appareil pour enregistrer. Le diagramme d'un système approprié d'analyse à
flux segmenté (SFS) est donné à l'annexe A (figure A.1). Noter que ces diagrammes peuvent avoir besoin d'être
adaptés en fonction du système utilisé.
6.3.2  Tube de réduction Cd/Cu
Tubes en verre en forme de U, d'environ 15 cm de longueur et de 2 mm de diamètre intérieur, dotés de ferrures
pour raccordement aux tubes du SFS. Ils peuvent être achetés remplis de cadmium auprès du fabricant du système
SFS, ou le remplissage peut être fait ultérieurement (voir 6.2.3 et 6.2.4).
NOTE —  Cet appareil SFS permet de déterminer le nitrite simplement en court-circuitant la colonne de réduction (voir
annexe A, figure A.2).
