This International Standard describes a method for the detection and quantification of N-nitrosodiethanolamine (NDELA) in cosmetics and raw materials used in cosmetics by high performance liquid chromatography (HPLC) coupled with post-column photolysis and derivatization. This method is not applicable to the detection and/or quantification of nitrosamines other than NDELA, nor to the detection and/or quantification of NDELA in products other than cosmetics or raw materials used in cosmetics. If a product has the possibility of either NDELA contamination from the ingredients or NDELA formation by the composition of ingredients, the method will be applied for the testing of cosmetic products and is an alternative to ISO 15819. This method is not applicable to matrices containing oxidation dyes.

Ta mednarodni standard opisuje metodo za detekcijo in količinsko določevanje n-nitrozodietanolamina (NDELA) v kozmetičnih izdelkih in surovinah, uporabljenih v kozmetičnih izdelkih, s tekočinsko kromatografijo visoke ločljivosti (HPLC), združeno s postopkom fotolize in derivacije za kolono. Ta metoda se ne uporablja za detekcijo in/ali količinsko določevanje N-nitrozaminov, ki niso NDELA, niti za detekcijo in/ali količinsko določevanje NDELA v proizvodih, ki niso kozmetični, oziroma surovinah, uporabljenih v kozmetičnih proizvodih. Če za proizvod obstaja možnost bodisi kontaminacije z NDELA zaradi sestavin bodisi tvorbe NDELA zaradi sestave sestavin, se ta metoda uporabi za preskušanje kozmetičnih izdelkov ter nadomešča ISO 15819. Введение
Воздействие N-нитрозаминов на человека может происходить из разных источников, таких как
окружающая среда, продукты питания и личной гигиены. Для сохранения здоровья человека, по
результатам воспринимаемого канцерогенного потенциала для некоторых видов животных, очень
важным признано сведение к минимуму воздействие N-нитрозаминов. Среди N-нитрозаминов, N-
нитрозодиэтаноламин (NDELA) признан как потенциальный загрязнитель косметических средств.
В данном контексте разработаны несколько аналитических методов для обнаружения и определения
присутствия NDELA в косметических средствах. Примерами этих методов являются газовая
хроматография/анализ тепловой энергии, жидкостная хроматография высокого разрешения в
сочетании либо с определением масс-спектрометрией, либо с фотолизом и колориметрической
квантификацией. В последнем методе используется специальная технология, обеспечивающая
особенность NDELA, с целью минимизировать риск искусственного образования вещества,
определяемого при анализе, и провести точное определение количества.
Данный аналитический метод использует жидкостную хроматографию высокого разрешения (HPLC) в
сочетании с пост-колоночным фотолизом и получением производных с целью разделить и выявить
уровни микроэлементов NDELA из косметических ингредиентов или матрицы продукта с особенностью
для NDELA.
Настоящий международный стандарт ссылается на совместное исследование (Ссылка [2]) семи
лабораторий, опубликованное в 2006. Критерий достоверности приведен в Ссылке [2].

1 Область применения
В настоящем международном стандарте описан метод обнаружения и определения количества
N-нитрозодиэтаноламина (NDELA) в косметических средствах и сырье, применяемом в косметике, с
помощью жидкостной хроматографии высокого разрешения (HPLC), пост-колоночным фотолизом и
получением производных.
Данный метод не применяется ни к обнаружению и/или определению количества нитрозаминов,
отличающихся от NDELA, ни к обнаружению и/или определению NDELA в продуктах, не относящихся к
косметике или к сырью, используемому в косметических средствах.
Если продукт имеет возможность либо загрязнения NDELA от ингредиентов, либо от образования
NDELA из состава ингредиентов, то метод применяется для тестирования косметических продуктов и
является альтернативой ISO 15819.
Данный метод не применяется к матрицам, содержащим окислительные красители.
2 Принцип
Извлечение нитрозамина NDELA из косметических проб выполняется с помощью воды. Очистка
выполняется либо с применением твердофазного извлечения (очистка SPE, см. 5.3.2) с картриджем
C18 либо с применением дихлорметана (очистка DCM, см. 5.3.3), если пробы не диспергируют в воде.
Экстракты анализируют методом HPLC, пост колоночным фотолизом и получением производных.
NDELA отделяют от косметической матрицы, применяя обратнофазную жидкостную хроматографию.
N-нитрозная связь разрывается УФ фотолизом с образованием иона нитрита. Согласно реакции
Грисса, нитритная функциональная группа диазотируется сульфаниламидом в кислой среде, а затем
соединяется с N-(1-нафтил) этилендиамин дигидрохлоридом (NED), образуя азокраситель пурпурного
цвета, который количественно определяется методом спектрофотометрии при максимальной длине
волны, λ , равной 540 нм (см. Приложение B).

Присутствие NDELA может подтверждаться повторением анализа без фотолиза (тогда нитритный ионн
не образуется, поскольку N-нитрозная связь не разорвана). Отсутствие хроматографического пика во
время удержания NDELA на хроматограмме подтверждает, что пик, наблюдаемый при первом анализе,
соответствует NDELA.
3 Реактивы
3.1 Метанол, с чистотой для HPLC.
3.2 Вода, с чистотой для HPLC.
3.3 Дихлорметан, с чистотой для HPLC.

3.4 N-Нитрозодиэтаноламин, с известной чистотой выше 95 %. CAS No. [116-57-7].
3.5 Ортофосфорная кислота, 85 %, аналитической чистоты.
3.6 Ацетат аммония, аналитической чистоты.
3.7 Раствор ацетата аммония, 1 моль/л.
Растворяют 77,08 г ацетата аммония (3.6) в 1,0 л воды (3.2).
3.8 Раствор ацетата аммония, 0,02 моль/л.
Берут 20 мл раствора 1 моль/л-ацетата аммония (3.7) и разбавляют до 1 л водой (3.2).
3.9 N-(1-Нафтил) этилендиамин дигидрохлорид, с известной чистотой выше 98 %. CAS No. [1465-25-4].
Этот реактив должен быть герметизирован и храниться в темноте.
3.10 Сульфаниламид, с известной чистотой выше 99 %. CAS No. [63-74-1].
3.11 Реактив Грисса.
Растворяют 0,25 г N-(1-нафтил) этилендиамин дигидрохлорида (3.9) в воде (3.2) и разбавляют до 250
мл в мерной колбе. Растворяют 4,0 г сульфаниламида (3.10) в 250 мл 5 %-ого (масса/объем) водного
раствора 85 %-ной ортофосфорной кислоты (3.5). Смешивают реактивы вместе в склянке из
янтарного стекла и держат смесь в темноте.
Смесь может использоваться в течение пяти дней и должна оставаться в любом случае бесцветной,
если она храниться при температуре от 2 °C до 8 °C.
4 Аппаратура
Используют стандартную лабораторную посуду и оборудование, а также следующее.

Introduction
Human exposure to N-nitrosamines can occur through diverse sources such as the environment, food or
personal care products. As a result of their perceived carcinogenic potential on several animal species,
minimization of exposure to N-nitrosamines is recognized as important to the preservation of human health.
Among N-nitrosamines, N-nitrosodiethanolamine (NDELA) has been recognized as a potential contaminant of
cosmetics.
In this context, several analytical methods have been developed to detect and determine the presence of
NDELA in cosmetics. Examples of these methods are gas chromatography/thermal energy analysis, and high
performance liquid chromatography coupled either with a mass spectrometry determination or with photolysis
and colorimetric quantification. The latter method uses specific technology to ensure specificity towards
NDELA, to minimize the risk of artefactual formation of the analyte of interest and to allow precise
quantification.
This analytical method uses High Performance Liquid Chromatography (HPLC) coupled with post-column
photolysis and derivatization, in order to separate and detect trace levels of NDELA from a cosmetic
ingredient or product matrix with specificity for NDELA.
This International Standard refers to a collaborative study (Reference [2]) involving seven laboratories and
published in 2006. Validation criteria are given in Reference [2].

1 Scope
This International Standard describes a method for the detection and quantification of
N-nitrosodiethanolamine (NDELA) in cosmetics and raw materials used in cosmetics by high performance
liquid chromatography (HPLC) coupled with post-column photolysis and derivatization.
This method is not applicable to the detection and/or quantification of nitrosamines other than NDELA, nor to
the detection and/or quantification of NDELA in products other than cosmetics or raw materials used in
cosmetics.
If a product has the possibility of either NDELA contamination from the ingredients or NDELA formation by the
composition of ingredients, the method will be applied for the testing of cosmetic products and is an alternative
to ISO 15819.
This method is not applicable to matrices containing oxidation dyes.
2 Principle
Extraction of the nitrosamine NDELA from cosmetic samples is carried out with water. Clean-up is performed
either using solid phase extraction (SPE clean-up, see 5.3.2) with a C18 cartridge or dichloromethane (DCM
clean-up, see 5.3.3) when the samples are not dispersible in water. The extracts are analysed by HPLC, post-
column photolysis and derivatization. NDELA is separated from the cosmetic matrix using reversed-phase
liquid chromatography. The N-nitroso bond is cleaved by UV photolysis with the formation of nitrite ion.
According to the Griess reaction, the nitrite functional group is diazotized with sulfanilamide in an acid medium
and is then coupled with N-(1-naphthyl)ethylenediamine dihydrochloride (NED) to form a purple-coloured azo
dye that is quantitatively determined spectrophotometrically at a maximum wavelength, λ , of 540 nm (see

Annex B).
The presence of NDELA can be confirmed by repeating the analysis without photolysis (no nitrite ion is then
produced because the N-nitroso bond is not cleaved). The absence of a chromatographic peak at the
retention time of NDELA in the chromatogram confirms that the peak observed in the first analysis
corresponds to NDELA.
3 Reagents
3.1 Methanol, HPLC grade.
3.2 Water, HPLC grade.
3.3 Dichloromethane, HPLC grade.
3.4 N-Nitrosodiethanolamine, with known purity greater than 95 %. CAS No. [116-57-7].

3.5 Orthophosphoric acid, 85 %, analytical grade.
3.6 Ammonium acetate, analytical grade.
3.7 Ammonium acetate solution, 1 mol/l.
Dissolve 77,08 g of ammonium acetate (3.6) in up to 1,0 l of water (3.2).
3.8 Ammonium acetate solution, 0,02 mol/l.
Take 20 ml of 1 mol/l ammonium acetate solution (3.7) and dilute to 1 l with water (3.2).
3.9 N-(1-Naphthyl) ethylenediamine dihydrochloride, with known purity greater than 98 %. CAS

3.11 Griess reagent.
Dissolve 0,25 g N-(1-naphthyl) ethylenediamine dihydrochloride (3.9) in water (3.2) and dilute to 250 ml in a
volumetric flask. Dissolve 4,0 g of sulfanilamide (3.10) in 250 ml of a 5 % (mass/volume) aqueous solution of
85 % orthophosphoric acid (3.5). Mix the reagents together in an amber glass bottle and keep the mixture
away from the light.
The mixture can be used for five days and shall, in any case, remain colourless when it is stored between 2 °C
and 8 °C.
4 Apparatus
Use standard laboratory glassware and equipment, and the following.

Introduction
Human exposure to N-nitrosamines can occur through diverse sources such as the environment, food or
personal care products. As a result of their perceived carcinogenic potential on several animal species,
minimization of exposure to N-nitrosamines is recognized as important to the preservation of human health.
Among N-nitrosamines, N-nitrosodiethanolamine (NDELA) has been recognized as a potential contaminant of
cosmetics.
In this context, several analytical methods have been developed to detect and determine the presence of
NDELA in cosmetics. Examples of these methods are gas chromatography/thermal energy analysis, and high
performance liquid chromatography coupled either with a mass spectrometry determination or with photolysis
and colorimetric quantification. The latter method uses specific technology to ensure specificity towards
NDELA, to minimize the risk of artefactual formation of the analyte of interest and to allow precise
quantification.
This analytical method uses High Performance Liquid Chromatography (HPLC) coupled with post-column
photolysis and derivatization, in order to separate and detect trace levels of NDELA from a cosmetic
ingredient or product matrix with specificity for NDELA.
This International Standard refers to a collaborative study (Reference [2]) involving seven laboratories and
published in 2006. Validation criteria are given in Reference [2].

1 Scope
This International Standard describes a method for the detection and quantification of
N-nitrosodiethanolamine (NDELA) in cosmetics and raw materials used in cosmetics by high performance
liquid chromatography (HPLC) coupled with post-column photolysis and derivatization.
This method is not applicable to the detection and/or quantification of nitrosamines other than NDELA, nor to
the detection and/or quantification of NDELA in products other than cosmetics or raw materials used in
cosmetics.
If a product has the possibility of either NDELA contamination from the ingredients or NDELA formation by the
composition of ingredients, the method will be applied for the testing of cosmetic products and is an alternative
to ISO 15819.
This method is not applicable to matrices containing oxidation dyes.
2 Principle
Extraction of the nitrosamine NDELA from cosmetic samples is carried out with water. Clean-up is performed
either using solid phase extraction (SPE clean-up, see 5.3.2) with a C18 cartridge or dichloromethane (DCM
clean-up, see 5.3.3) when the samples are not dispersible in water. The extracts are analysed by HPLC, post-
column photolysis and derivatization. NDELA is separated from the cosmetic matrix using reversed-phase
liquid chromatography. The N-nitroso bond is cleaved by UV photolysis with the formation of nitrite ion.
According to the Griess reaction, the nitrite functional group is diazotized with sulfanilamide in an acid medium
and is then coupled with N-(1-naphthyl)ethylenediamine dihydrochloride (NED) to form a purple-coloured azo
dye that is quantitatively determined spectrophotometrically at a maximum wavelength, λ , of 540 nm (see
max
Annex B).
The presence of NDELA can be confirmed by repeating the analysis without photolysis (no nitrite ion is then
produced because the N-nitroso bond is not cleaved). The absence of a chromatographic peak at the
retention time of NDELA in the chromatogram confirms that the peak observed in the first analysis
corresponds to NDELA.
3 Reagents
3.1 Methanol, HPLC grade.
3.2 Water, HPLC grade.
3.3 Dichloromethane, HPLC grade.
3.4 N-Nitrosodiethanolamine, with known purity greater than 95 %. CAS No. [116-57-7].

3.5 Orthophosphoric acid, 85 %, analytical grade.
3.6 Ammonium acetate, analytical grade.
3.7 Ammonium acetate solution, 1 mol/l.
Dissolve 77,08 g of ammonium acetate (3.6) in up to 1,0 l of water (3.2).
3.8 Ammonium acetate solution, 0,02 mol/l.
Take 20 ml of 1 mol/l ammonium acetate solution (3.7) and dilute to 1 l with water (3.2).
3.9 N-(1-Naphthyl) ethylenediamine dihydrochloride, with known purity greater than 98 %. CAS
No. [1465-25-4].
This reagent should be sealed and stored in the absence of light.
3.10 Sulfanilamide, with known purity greater than 99 %. CAS No. [63-74-1].
3.11 Griess reagent.
Dissolve 0,25 g N-(1-naphthyl) ethylenediamine dihydrochloride (3.9) in water (3.2) and dilute to 250 ml in a

Introduction
L'exposition humaine aux N-nitrosamines peut se produire au contact de sources diverses présentes dans
l'environnement, la nourriture ou les produits de soin personnels. Ces substances ayant montré un effet
cancérigène notoire sur plusieurs espèces animales, il est reconnu qu'une restriction d'exposition aux
N-nitrosamines est d'une importance primordiale pour la préservation de la santé humaine. Parmi les
N-nitrosamines, il a été établi que la N-nitrosodiéthanolamine (NDELA) est un contaminant potentiel des
cosmétiques.
Dans ce contexte, plusieurs méthodes analytiques ont été développées pour rechercher et doser sa présence
dans les cosmétiques, comme la chromatographie en phase gazeuse couplée à l'analyse d'énergie
thermique (TEA), la chromatographie en phase liquide à haute performance (CLHP) couplée, soit à une
détermination par spectrométrie de masse, soit à une photolyse avec quantification colorimétrique. Cette
dernière méthode nécessite l'usage d'une technologie particulière dans le but de garantir une sélectivité
maximale lors de la recherche de la NDELA, afin de limiter le plus possible le risque de formation artefactuelle
de l'analyte recherché et de permettre une quantification précise de la substance.
La présente méthode analytique emploie la chromatographie en phase liquide à haute performance (CLHP)
associée à une photolyse et à une dérivation post-colonne afin de séparer et de rechercher les traces de
NDELA dans un ingrédient cosmétique ou dans une matrice de produit présentant une sélectivité à la NDELA.
La présente Norme internationale fait référence à une étude menée en collaboration par sept laboratoires
(Référence [2]) et publiée en 2006. Des critères de validation sont donnés dans la Référence [2].

1 Domaine d'application
La présente Norme internationale décrit une méthode de recherche et de quantification de la NDELA dans les
cosmétiques et les matières premières utilisées dans les cosmétiques par CLHP, photolyse et dérivation post-
colonne.
La présente méthode ne s'applique pas à la recherche et/ou à la quantification des nitrosamines autres que la
NDELA, ni à la recherche et/ou à la quantification de la NDELA dans les produits autres que les cosmétiques
ou les matières premières utilisées dans les cosmétiques.
Si un produit est susceptible d'être contaminé par la NDELA contenue dans ses ingrédients ou que la
formation de NDELA est possible du fait de la composition en ingrédients, alors la méthode est à appliquer
lors des essais des produits cosmétiques et constitue une alternative à l'ISO 15819.
Le présente méthode ne s'applique pas aux matrices contenant des colorants d'oxydation.
2 Principe
L'extraction de la nitrosamine NDELA des échantillons cosmétiques est réalisée avec de l'eau. La purification
est effectuée soit par extraction en phase solide (purification SPE, voir 5.3.2) à l'aide d'une cartouche C18,
soit à l'aide de dichlorométhane (purification DCM, voir 5.3.3) lorsque les échantillons ne peuvent pas se
disperser dans l'eau. Les extraits sont analysés par CLHP, photolyse et dérivation post-colonne. La NDELA
est séparée de la matrice cosmétique par chromatographie en phase liquide inverse. La liaison N-nitroso est
coupée par photolyse UV avec formation d'ion nitrite. Selon la réaction de Griess, le groupe fonctionnel nitrite
est diazoté par du sulfanilamide en milieu acide puis associé à du chlorhydrate de N-(1-naphthyl)éthylène
diamine (NED) pour former un colorant azoïque pourpre qui est ensuite dosé quantitativement par
spectrophotométrie à λ 540 nm (voir Annexe B).

La présence de NDELA peut être confirmée en procédant à une nouvelle analyse sans photolyse (aucun ion
nitrite n'est alors produit car la liaison N-nitroso n'est pas coupée). L'absence de pic chromatographique au
temps de rétention de la NDELA sur le chromatogramme confirme que le pic observé lors de la première
analyse correspond bien à la NDELA.

3.5 Acide orthophosphorique 85 %, de qualité analytique.
3.6 Acétate d'ammonium, de qualité analytique.
3.7 Solution d'acétate d'ammonium, à 1 mol/l.
Dissoudre 77,08 g d'acétate d'ammoniun (3.6) dans 1,0 l d'eau (3.2).
3.8 Solution d'acétate d'ammonium, à 0,02 mol/l.
Prélever 20 ml de solution d'acétate d'ammonium 1 mol/l (3.7) et compléter à 1 l avec de l'eau (3.2).
3.9 Chlorhydrate de N-(1-naphthyl)éthylène diamine, de pureté connue supérieure à 98 %.

Il convient que ce réactif soit scellé et conservé hermétiquement à l'abri de la lumière.
3.10 Sulfanilamide, de pureté connue supérieure à 99 %. Numéro CAS: [63-74-1].
3.11 Réactif de Griess.
Dissoudre, dans une fiole jaugée, 0,25 g de chlorhydrate de N-(1-naphthyl)éthylène diamine (3.9) dans de
l'eau (3.2) et compléter à 250 ml. Dissoudre 4,0 g de sulfanilamide (3.10) dans 250 ml d'une solution aqueuse
à 5 % (masse/volume) d'acide orthophosphorique 85 % (3.5). Mélanger les réactifs dans un flacon en verre
ambré et conserver le mélange à l'abri de la lumière.
Le mélange peut être utilisé pendant 5 jours et doit, en tout cas, rester incolore lorsqu'il est conservé à une
température comprise entre 2 °C et 8 °C.
4 Appareillage
Utiliser une verrerie et un matériel de laboratoire normalisés, ainsi que l'équipement suivant.
